Virus titration of melanoma-specific TCRs in the Jurkat/MA cell line (PDF, 52.9 KB) - Transient viral supernatant of vectors encoding the gene-optimized Mart-1-specific TCRs DMF4 (▲), 1D3 (●), or 2C2 (■), and Gp100-specific TCR R6C12 (♦), was generated. The indicated amounts of viral supernatant were supplemented with 5% IMDM medium to a total volume of 1 mL, and used to transduce Jurkat/MA cells. Transduced cells were stained with anti-TCR 4 days after transduction. The percentage of TCR+ cells is depicted. The lines indicate the dilutions of viral supernatant that result in comparable expression of the different TCRs and that were used in further experiments. Bars represent range. Figure S2.
