Sorafenib, but not sunitinib, affects function of dendritic cells and induction of primary immune responses

  • Figure S1. Sorafenib lowers expression of cell surface molecules on DCs stimulated with TLR3 ligand (JPG, 193 KB) -
    DCs were generated by incubating adherent monocytes with GM-CSF and IL-4. Poly I;C was added as a stimulus to the culture medium. Mature DCs were exposed for 24 hours to sunitinib (A) or sorafenib at the indicated concentrations (B). The changes in phenotype of DCs were analyzed by flow cytometry after staining with Abs against CD40, CD80, CD86, CD83 or.CCR7 Shaded histograms represent isotype control and open diagrams staining with the specific antibody. The numbers represent mean fluorescence intensity. One representative experiment out of at least three is shown.





  • Figure S2. TL3 ligand-induced DC migration toward CCL19/MIP-3β is impaired by sorafenib (JPG, 15.5 KB) -
    DCs were generated by incubating adherent monocytes with GM-CSF and IL-4. Poly I;C was added as a stimulus to the culture media. Mature DC were exposed for 24 hours to sorafenib at the indicated concentrations. Migration toward CCL19 was analyzed using transwell chambers. DCs (1 × 105) were seeded in the upper chamber in triplicates, and the number of migrated DCs was analyzed after 3 hours by counting gated DCs for one minute by FACS analysis (* P < 0.05; ** P < 0.001).





  • Figure S3. Sorafenib reduces lymphocyte stimulation capacity of DCs (JPG, 22.6 KB) -
    DCs were generated by incubating adherent monocytes with GM-CSF and IL-4 and treated with sorefenib for 24 hours. Poly I;C was added as a stimulus to the culture media. DCs (104 cells per well) were irradiated with 30 Gy and were incubated for five days with 105 allogenic PBMCs. Thymidine incorporation was measured by a 16-hour pulse with 3Hthymidine. One representative experiment out of four is shown.





  • Figure S4. Dose response to DC stimulation (JPG, 54.5 KB) -
    DCs were generated by incubating adherent monocytes with GM-CSF and IL-4 and treated with sunitinib for 24 hours. LPS was added as a stimulus to the culture media. Decreasing numbers of DCs (104, 103, 102 per well) were incubated for five days with 105 allogenic PBMCs. Thymidine incorporation was measured by a 16-hour pulse with 3Hthymidine.
