The kinetics of HLA-class I re-expression in the presence or absence of bortezomib after acid stripping (JPG, 24.9 KB) - JJN3 was exposed to an acid buffer (pH 2.5) for 3 min at 37°C to remove HLA from cell surface. Cells post acid stripping were re-cultured in the presence or absence of 1 µM bortezomib and incubated for indicated times at 37°C. PI negative cells were analyzed for HLA-class I expression. The data represent one of 3 individual experiments. The viability of cells post-acid treatment was 67 ± 4%, compared to untreated cells (96 ± 2%). The survival of JJN3 in the presence of this drug was 56-66%. The percentage of viable cells (67-70%) was similar in the absence of bortezomib (data not shown).
