Transcripts present in the MVs analysed by microarray were defined as those characterized by a positive linear relation between the transcript signal detected by the microarray analysis and the amount of total RNA hybridized (XLS, 113 KB) - This analysis was done hybridizing arrays with labeled-cRNA produced using three different concentrations of total RNA extracted from two independent preparations of MVs. A total of 298 transcripts (sheet 3) were found with this procedure, 183 were associated to RefSeq identifiers (sheet 1) and the remaining were Unigene EST (sheet 2). Figure S1. Equal amount of MVs were used to extract total RNA with or without RNase treatment by mean of TRIZOL method (JPG, 12.6 KB) - One µg of total RNA extracted from MVs and from MV-producing EPCs were labelled by oligo dT driven retrotranscription. For total RNA from RNase treated MVs a volume equal to that of the untreated MVs corresponding to 1 µg of total RNA was labelled. Labelling conditions: dNTP(-dATP) 25 mM; unlabeled dATP 120 µM; 32P-ATP(alpha) 10µCi (3000 Ci/mmol). Retrotranscription was performed, in final volume of 25 µl, for 1 hour at room temperature using the High-Capacity cDNA Kit (Applied Biosystems). Five µl of each reaction were loaded on a 0.6% Agarose gel in TBE 0.5X. After drying the gel was exposed at room temperature for 72 hours. Figure S2.
