After 20 days of coculture of CD34+ CB cells on OP9-DL1 cells in the presence of Flt-3L, SCF, IL-7, and IL-15, cells were sorted according to CD56 expression (start) and approximately 20 × 103 to 30 × 103 sorted cells were recultured on OP9-DL1 for different time points, as indicated. Note the appearance and progressive increase in CD56 expression in the sorted CD56− and CD56dim subsets, respectively, whereas the CD56high subset remains CD56high. Histograms are representative of two independent experiments.

Cells were obtained after 22 days of coculture of CD34+ CB cells on OP9-DL1 or OP9-control stromal cells with Flt-3L, SCF; IL-7, and IL-15. The results show mean plus or minus SD of two independent experiments.
