Comparison of cultured bone marrow (BM) and circulating peripheral blood (PB) CD34+ cells from a single subject with ET (JPG, 99.4 KB) - (A) Immunophenotypic analysis of erythroid markers CD71 and Glycophorin A from days 2 through 14 of culture demonstrate a modest increase in early erythroid cells (gated as group 1) in BM compared to PB early (days 2 and 5) but comparable day 7 and beyond. Extracts prepared from HEL92.1.7 cells, which are homozygous for the V617F mutation, served as controls. Genotyping of the V617F mutation was performed in mononuclear cells or granulocytes by allele-specific PCR and BstXI restriction analysis.
