293T cells were cotransfected with SOCS-1 and either v-Abl or Pim-1, and treated with 4 µM of imatinib for 16 hours as labeled for each lane. Total lysates were analyzed by Western blot. Phosphorylation of SOCS-1 by v-Abl or Pim-1 results in mobility shift of SOCS-1 as previously described (P. Chen et al. 2002, PNAS. 99:2175-2180).

Immunofluorescence staining was performed. Images were acquired using a confocal microscope (model LSM-510; Carl Zeiss MicroImaging, Inc.) and a 63× objective (Carl Zeiss MicroImaging, Inc.). (A) V-Abl-transformed WT cells were labeled using indicated antibodies. SOCS-1 is located in the cytoplasm. Pim-1 protein is seen in both the cytoplasm and nucleus. The merged images of SOCS-1 (red) and Pim-1 (green) are shown as a yellow color. Bar, 10 µM. (B) V-Abl-transformed TKO cells were labeled with antibodies as indicated. The merged images (yellow) of SOCS-1 (red) and -tubulin (green) show that SOCS-1 colocalizes with the microtubule organizing complex (arrows), suggesting that SOCS-1 is mainly located in MTOC-associated proteasome as described in our previous studies (Vuong et al., Mol Cell Biol., 2004, 24: 9092-9101).
