C57Bl/6 mice were given an intraperitoneal injection of 300 µg control antibody or anti-GITR antibody and were immunized with NP-KLH in alum 30 minutes later. 10 days later mice were bled and anti-NP specific IgM (A), IgG2a (B), and IgG1 (C) were measured from the serum.

24 hours after cecal ligation and puncture (CLP) or sham surgery, splenocytes were harvested and flow cytometry was performed. Left panels show representative examples examining A) CD28, B) CTLA-4, C) CD69 and D) GITR expression on living (Sytox Blue−) CD3+CD4+ T cells from sham (black histogram) or CLP (grey histogram). Right panels show the mean fluorescent intensity (MFI) of CD28, CTLA-4, or GITR expression, or the percentage of CD69+ TH cells from sham (n=3) or CLP (n=4) mice. * p < 0.05 by Student’s t test.
