Ly10 cells were exposed to DMSO or 20 µM AG490 for 20 h before BrdU was added to the culture medium to a final concentration of 10 µM. Cells were cultured for another 90 minutes before fixation, staining with FITC-labeled anti-BrdU antibody and, finally, propidium iodide for 30 minutes. Numbers in the boxed areas in the FACS profile indicate the percentages of the BrdU positive S-phase fraction, sub-G1 fraction that is BrdU-positive and BrdU-negative, respectively.
