IL-21–mediated Foxp3 suppression leads to enhanced generation of antigen-specific CD8+ cytotoxic T lymphocytes

    Antigen-specific CTL generated from cultures initiated with CD25-depleted PBMC and treated with IL-21 were evaluated for recognition of endogenous target antigen expressed by tumor cells. Effector cells from cultures represented in Figures 1 and 2 were used in a chromium release assay at an effector to target ratio of 20:1 (Methods: 5,000 target cells labeled for 1 hour with Cr51 were co-cultivated with effector CTL at an E:T of 20:1 for 4 hours. Released Cr51 was measured with a gamma scintillation counter and percent specific lysis was determined by using the formula: percent specific release = experimental release – spontaneous release/total release. Spontaneous release was <10% of the total release in all assays). 375 is an A2+, MART-1+, NY-ESO-neg tumor cell line. 526 is an A2+, MART-1-neg, NYESO+ tumor cell line. OV16 is an A2+, WT-1+, MART-1-neg, NY-ESO-1-neg ovarian tumor cell line. For MART-1-specific T cell responses, specific lysis is seen against the Ag+ Tumor (526), but not the Ag-negative tumor line (375). Similarly for cultures generated using NY-ESO-1 and WT-1 peptides, specific lysis is seen against their respective Ag+ tumor targets.
