Loss of Bim leads to an abnormally increased number of memory B cells in peripheral blood (JPG, 70.9 KB) - Wt and bim−/− mice were immunized with NP coupled to KLH and 14 days later peripheral blood leukocytes were analyzed by immunofluorescent staining and flow cytometric analysis (as described in Figure 1A). Live cells were gated on dump-channel negative (IgM−IgD−Gr-1−Mac-1−) and B220+ cells (R1). Cells in R1 with a switched IgG1 isotype and the ability to bind the immunizing hapten NP coupled to allophycocyanin reside within the population of antigen-specific IgG1+NP+ B cells (R2). Amongst these (R1,R2) cells, the level of CD38 expression was determined to discriminate between memory (CD38high) and GC B cells (CD38low).
