Tube formation of HDMECs transfected with empty vector (Evec), CEACAM1 (CC1), mutated CEACAM1 (CC1/y--), CEACAM1-siRNA (CC1-siRNA) and Luciferase-siRNA after stimulation with VEGF and VEGF-C. (PDF, 207 KB) - The stimulation of empty vector transfected HDMECs (Evec) with both VEGF and VEGF-C induces endothelial tube formation (arrows) as expected. The length and the network of tubes (arrows) are markedly increased when stimulating CEACAM1 (CC1)-overexpressing HDMECs with VEGF and VEGF-C whereat the strongest tube formation effect is achieved after stimulation with VEGF-C. This effect is almost completely abolished when HDMECs overexpressing mutated CEACAM1 (CC1/y--) are stimulated with VEGF or VEGF-C. Only few tubes are seen when CECAM1-silenced (CC1-siRNA) HDMECs are stimulated with VEGF and VEGF-C while luciferase-silenced (Luci-siRNA) HDMECs form tubes after stimulation with VEGF and VEGF-C comparable to empty vector transfected HDMECs. Figure S2. Increased expression of LYVE-1 and Prox1 by CEACAM1 overexpression in HDMECs. (PDF, 76 KB) - CEACAM1-overexpressing HDMECs CEACAM1(+) also exhibit an increased staining for Prox1 (A) and LYVE-1 (C) when compared with the corresponding controls transfected with empty vector as shown in panels B and D. Note that in panel A some cells exhibit a strong nuclear or perinuclear staining (arrows) while the others show a cytoplamsic staining for Prox1. Cells are counterstained with Calcium Red (red staining of the cell nuclei).
