Polymerization of plasma clots was imaged in real time by using a Delta Vision Spectris Restoration microscopy system equipped with an Olympus IX–70 inverted microscope with a CCD camera. Epi-fluorescence illumination was provided by a HBO – 100 W mercury arc lamp and the excitation wavelength was selected with an electronic filterwheel containing a filter for Alexa 488, dichromatic mirror and emission filter for Alexa 488. Plasma clots were prepared by mixing pooled plasma with tissue factor and CaCl2 as described in detail in the Materials and Methods section. Examples are shown of two experiments at different magnifications. While Fig. 1 of the paper shows just a few time points during polymerization, the videos show the polymerization process in more detail over the entire time course.
