(A) Reconstituted primitive erythroid islands are easily identified by y globin-positive primitive erythroblasts attached to an F4/80-positive macrophage. (B) Occasional unstripped islands (D) found usually near the edge of the slide chambers, and are easily distinguished by the large numbers of y globin-negative definitive cells with larger nuclei. Single macrophages (m in B) as well as F4/80-negative cells (C) are also identifiable based on differential F4/80 antibody and HO staining. Figure S2. Primitive erythroid cells do not spontaneously enucleate when cultured in vitro using conditions used for erythroblast island reconstitution (JPG, 26.1 KB) - No significant increase in primitive erythroid cell enucleation was seen in control experiments when fetal blood was incubated for 24 hours in the absence of adherent fetal liver cells. Mean +/− SEM percent enucleation at 0 and 24 hours for 3 independent experiments are shown.
