Platelet rich plasma (PRP), mononuclear cell (MNC) and granulocyte (GRC) fractions along with the red blood cells (RBC) were obtained. Immunopurified monocytes were nucleofected, and the whole blood was reconstituted from PRP, RBC, lymphocytes and granulocytes depleted for residual monocytes. Upon completion of experimental treatments, MNC or granulocyte fractions were isolated and assayed.

Platelet rich plasma (PRP), mononuclear cell (MNC) and granulocyte (GRC) fractions, and red blood cells (RBC) were obtained. Granulocytes, depleted for monocyte contamination, were nucleofected and the whole blood was reconstituted using PRP, MNC fraction, and RBC. Upon completion of experimental treatments, MNC or granulocyte fractions were again isolated and assayed.
