Comparison of 51Cr release and flow cytometry based cytotoxicity assays (JPG, 32.7 KB) - Day 0 and day 20 cells from patients 1, 2, 4, and 5 used as effectors against the K562 cell line to document the reliability of flow cytometry based cytotoxicity assay compared to the standard 51Cr release assay. Figure S2. Cytotoxicity of day 0 and day 20 NK cells from MM patients against autologous tumor cells (JPG, 59 KB) - BM samples were stained with TFL4 and co-incubated with effector cells for four hours at an E:T ratio of 1:10. Prior to acquisition by flow cytometry, the samples were stained with mAbs against CD38 and CD138 as well as the viability dye 7-AAD. During the analysis of the flow cytometry data, BM cells were gated as the TFL4+ population. Within this gate, MM cells were identified as CD38+CD138+. The histograms show the percentage of live/dead cells (using 7-AAD) in this population at the end of the assay. The data for patient 5 are presented in Figure 3c.
