FANCM is phosphorylated throughout the cell cycle (JPG, 68.1 KB) - HeLa cells synchronized as described in Figure 2A were collected at the indicated time points after release from G1/S boundary (lines 1-7) and M phase (lanes 8-12), respectively. The chromatin fractions separated from soluble fractions were incubated with phosphatase and then analyzed by western blotting. The SDS-PAGE gel used here was 3-8% Tris-Acetate gel. Figure S2. FAAP24 depletion in HeLa cells displays hypersensitivity to MMC treatment (JPG, 60.6 KB) - (A) HeLa cells were transfected with the indicated siRNAs for 48 h, and then exposed to the indicated concentration of MMC for 5 days in 96 well plates. Cell survival was determined by CyQuant cell proliferation assay kit. (B) HeLa cells treated as in (A) were exposed to MMC for 5 days in 6 well plates. Cell viability was determined by trypan blue exclusion.
