Human mononuclear cells were incubated with either test antibody or isotype control prior to washing and transplant into mice. After six weeks the percentage of human cells in the bone marrow was determined. Horizontal bars represent mean engraftment percentage. 5 million cord blood mononuclear cells were injected per mouse. There was a significant difference between control and anti-CD34 antibody (P<0.001) and anti-CD45 antibody (P<0.001).

Cord blood mononuclear cells were stained with PKH26 and then incubated with isotype control or anti-CD38 antibody (either HIT2 or HB7 clones). After washing, 1.9 million cells were transplanted via intravenous route into each mice. At sixteen hours mice were sacrificed and bone marrow underwent analysis by flow cytometry. The number of PKH26 bright cells was determined after acquisition of more than 5 × 105 events. An example is shown from control (A) and anti-CD38 antibody (B) groups. The region marks the PKH26 bright cells. (C) Both clones of anti-CD38 antibody tested reduced the number of cells in the bone marrow significantly at sixteen hours (P<0.001).
