    Conditioned medium from LPS-challenged CSF−/−/GM-CSF−/− MEFs was partially purified by a series of chromatographic steps. Details for each chromatographic procedure are presented in Methods.

    The protein profile of each chromatographic step, determined by absorbance at 280nM, is shown as a continuous line. Fractions from each chromatographic run were pooled pair-wise and tested for NPA as described in Methods. NPA content is shown by shaded bars. (A) Blue Sepharose depletion of serum albumin from CSF−/−/GM-CSF−/− MEFs conditioned medium. (B) Size exclusion chromatography on Superose 12. Biological assay of paired fractions showed two pools of activity: Pool A fractions 31-40; Pool B fractions 41-62. (C) Anion exchange chromatography of Pool A on Mono Q.
