Targeting Bcl-2 family proteins modulates the sensitivity of B-cell lymphoma to rituximab-induced apoptosis
Blood Stolz et al. 112: 3312

Supplemental materials for: Stolz et al

Files in this Data Supplement:

  • Figure S1. Isotype controls (JPG, 45.3 KB) -
    (A) Human B-NHL cell lines were incubated with medium, Rituximab or the isotype control and mononuclear cells. The CD20-negative human leukemia cell line K562 served as negative control. Cell death was quantified flow cytometrically following staining with PI; mean values + SD of three independent experiments are shown. (B) Human B-NHL cell lines were incubated with medium, Rituximab or the isotype control and human serum. The CD20-negative human leukemia cell line K562 served as negative control. Cell death was quantified flow cytometrically following staining with PI; mean values + SD of three independent experiments are shown. (C) Flow cytometric analysis of CD20 expression of SU-DHL-4 and Ramos B-NHL cells transduced with control vector or a Bcl-xL expression vector. Note that transgenic expression of Bcl-xL fails to downmodulate endogenous CD20 expression.

  • Figure S2. Specificity controls for lymphoma xenograft models (JPG, 33.5 KB) -
Kaplan-Meier plots of symptom-free survival of NOD/SCID mice following intravenous inoculation of 107 Ramos cells or HT cells. Starting on day 5, mice received intraperitoneal injections of isotype control (green) or vehicle (grey).

  • Figure S3. Activity of ABT-737 alone and in combination with Rituximab in B-NHL cells (JPG, 37.8 KB) -
    (A) Human B-NHL cell lines were incubated with increasing concentrations of the BH3-mimetic ABT-737. Cell death was quantified flow cytometrically following staining with PI; mean values + SD of three independent experiments are shown. (B) Three Rituximab-sensitive human B-NHL cell lines were incubated with cross-linked Rituximab (Rc), ABT-737 or a combination of Rc and ABT-737. Cell death was quantified flow cytometrically following staining with PI; mean values + SD of three independent experiments are shown. Note that the addition of ABT-737 did not further enhance Rc-induced apoptosis. Note that the PTEN-deficient B-NHL cell lines Jeko-1 and HT were resistant to ABT-737–induced apoptosis.
