- Table S1. PCR and pyrosequencing primers (PDF, 304 KB) -
I, step 1 PCR primers; II, step 2 PCR primers; S, pyrosequencing primer, TSS, transcriptional start side.
- Table S2. Correlation of methylation results of investigated genes in the AML patients at diagnosis (PDF, 166 KB) -
Spearman nonparametric test. Correlation coefficients are shown in upper lines, P values in lower lines for each gene. Significant positive correlations are highlighted in orange, significant negative correlations in blue.
- Table S3. Genes hypermethylated at diagnosis in 3 or 4 AML patients (PDF, 19.1 KB)
- Table S4. Genes hypermethylated in relapse in 3 or 4 AML patients (PDF, 27.6 KB)
- Table S5. Genes hypermethylated at diagnosis and relapse in 3 or 4 AML patients (PDF, 14.4 KB)
- Table S6. Comparison of MCAM with bisulfite pyrosequencing (PDF, 11.4 KB) -
Results of methylation of HIN1, NOR1, NPM2, OLIG2, and PGR at diagnosis and relapse were compared in 4 AML patients.
- Figure S1. Maps of CpG islands of investigated genes (JPG, 100 KB)
-
Exons are shown as boxes: narrow boxes for untranslated and wide boxes for translated regions. Arrows indicate transcription start sites. Vertical bars above the x axes denote CpG sites. Boxes below the x axes show regions interrogated by pyrosequencing.

- Figure S2. Pyrograms (JPG, 714 KB)
-
Examples of pyrograms are shown for low and high methylation densities in the investigated genes.

- Figure S3. DNA methylation of studied genes in leukemia cell lines (JPG, 80.8 KB)
-
Cell lines are arranged horizontally. BV173R to TF1 are myeloid, CEM to Peer are T-cell, and ALL1 to RS4;11 are B-cell leukemia lines. Genes are arranged vertically. Mean methylation densities of 0%-10% are shown in white, 10%-50% methylation in gray, and more than 50% in black. DEL means homozygous deletion of the gene. All cell lines showed hypermethylation in most investigated genes. WBC, normal white blood cell controls showed methylation in LINE-1 only. SssI, in vitro methylated normal blood control DNA was hypermethylated in all studied genes.
