Supernatants of the indicated cell lines were analyzed by an IL-21 specific ELISA. The amount of IL-21 in the supernatants is shown in pg/ml. As controls, standard medium and the reagent diluent for the standard (reagent D) were included. Error bars denote SDs.

(A) L1236 and HDLM-2 cells were cultured for 12 hours in medium containing 0.5% FCS. Thereafter, cells were stimulated with 100 ng/ml rhIL-21, and whole cell extracts were prepared at the indicated times and analyzed by WB for pSTAT3 (upper panel) and, as a control, STAT3 expression (center), and by EMSA for STAT3 DNA binding activity (lower panel). (B) Whole cell extracts of various HRS cell lines were prepared and analyzed for pSTAT3 and STAT3 by WB. (C) L1236 and HDLM-2 cells were treated as described in (A). At the indicated times, mRNA was prepared and analyzed by RT-PCR for expression of IL6, MCL1, and, as a control, GAPDH.
