    (A) Nucleotide sequence analyses of JAK3 in CMK and CMY cell lines. (B) Tyrosine phosphorylation of STAT5 in starved AMKL cell lines. Cellular extracts from MO7e starved and then stimulated by GM-CSF were used as control. (C) Tyrosine phosphorylation of JAK3 was assessed by immunoprecipitation of total cellular extracts from starved cells. (D) CMK and CMY growth is sensible to JAK. Cell survival analysis of MEG-01, CMK, and CMY cells at 72 hours after incubation with increasing amount of JAK inhibitor I (Calbiochem/Merk, Nottingham, UK).

    (A) Cytological analyses of blood smears (HES) showed a massive expansion of myeloid compartment. (B) Histological sections of engrafted mice showed a diffuse proliferation of myeloid forms in spleen (panel a) and liver (panel b). (C) Analysis of spleen sections demonstrated an increased fibrosis by reticulin staining in MPLT487A- and MPLW515L-expressing mice, but not in MPLWT-engrafted mice. Histopathology of tissues was assessed by microscopic examination of paraffin-embedded tissues.
