PE-conjugated anti–Gr-1 mAb was used to stain neutrophils, and APC-conjugated anti-CD45 mAb to stain all leukocytes, such that neutrophils appear magenta. Both neutrophils (Gr-1+, CD45+; magenta) and other leukocytes (Gr-1−, CD45+ blue) can be seen undergoing rolling interactions in the venule, whereas only neutrophils are arrested on the endothelial surface. AlexaFluor 488-conjugated isolectin GS-IB4 from Griffonia simplicifolia was used to label the microvasculature, strongly staining adjacent capillaries, and adjacent muscle fibres are visible via autofluorescence.
