  • Figure S1. FcγRIIa mediates outside in signaling during clot retraction (JPG, 40.4 KB) -
    Washed platelets were pre-incubated with 20 µg/ml mAb IV.3 or an isotype control antibody and added to buffer containing 1 mM CaCl2 and 150 µg/ml soluble fibrinogen. Clot formation was initiated by addition of 0.1U/ml thrombin, and allowed to proceed for 15 minutes at 37°C. Note that mAb IV.3 inhibits clot retraction.

    Platelets from a variant Glanzmann thrombasthenic patient harboring a homozygous mutation in the codon encoding amino acid 724 of the integrin β3 cytoplasmic domain (Δ724) express normal levels of the αIIbβ3 complex and FcγRIIa, as reported by the binding of the αIIbβ3 and FcγRIIa specific mAbs AP2 and IV.3, respectively.
