(A) Expression of TRAIL receptors in TF-1, HeL and K562 cell lines. Cells were stained with specific mAbs anti TRAIL-R1, -R2, -R3, -R4, as described in Materials and Methods. Specific fluorescence histograms (grey) are superimposed to negative controls (isotype-matched irrelevant mAbs, empty histograms). A representative of 3 independent experiments is shown. (B) Flow cytometric analysis of TRAIL-induced apoptosis of TF-1 cells treated for 24 hrs with the indicated doses of TRAIL. Apoptosis was detected staining cells with Anexin V-FITC and PI. A representative of 5 independent experiments is shown. (C) Apoptosis of TF-1 cells treated with increasing doses of TRAIL for 24 hrs. The maximal cell death was obtained with the 100 ng/ml dose. Data from 5 independent experiments are expressed as means ± S.D.
