(A) ELISPOT assay after short-term expansion. Freshly thawed PBMCs from patient 21 were assayed by IFN-γ ELISPOT assay directly (top row) or after 1 week of expansion in the presence of 0.1 µM of PRAME peptide. The day 1 assay was performed using 200,000 cells/well, and the day 7 assay was performed using the expanded cells derived from 67,000 of freshly thawed PBMCs/well. Each well represents one of 5 replicate wells. The first number below each image represents the raw number of spots counted in the displayed well and the number in parentheses represents the number of peptide-specific spots/million freshly thawed PBMCs calculated using data from all 5 replicates. Neg indicates the number of spots in this set of wells did not meet criteria for significant difference from control wells. (B) The top panel illustrates the frequency of IFN-γ responding CD8+ T cells prior to expansion and the bottom panel represents the frequencies of IFN-γ responding CD8+ T cells following 3 × weekly stimulation with test peptide.
