Activation of αIIb transcription during megakaryocytic induction occurs at the level of elongation rather than initiation (JPG, 55.8 KB) - K562 cells treated 48 hours with 25 nM TPA or DMSO solvent control underwent chromatin immunoprecipitation (ChIP) with antibody to RNA polymerase II (α-RPII) or with control IgG. PCR primers were directed to αIIb upstream region, proximal promoter, and exon 13, as indicated. Templates included water control (H2O), input chromatin consisting of 0.1% of material used for immunoprecipitation, and ChIP products. PCR products separated on a 2% agarose gel were visualized with ethidium bromide staining. Weight marker (WM) bands shown are 194-310 base pair fragments of ΦX174-Hae III. Figure S2. Cooperative effects of Cdk9 inhibition and GATA-1 deficiency on in vivo expression of a target gene (JPG, 67.6 KB) - Following treatment for 9 days with 5 mg/kg/day IP Flavopiridol, mice were euthanized for immunohistochemical staining of spleens for Von Willebrand Factor. All samples were processed simultaneously under identical conditions. Comparisons include C57BL/6 (Wt) and GATA-1Lo (G1Lo) strains treated either with saline or Flavopiridol (FP). Wt mice treated with saline and FP showed identical staining intensities. Shown are representative immunoperoxidase stained splenic sections at 400× magnification.
