Effective elimination of fludarabine-resistant CLL cells by PEITC through a redox-mediated mechanism

    Primary normal lymphocytes and CLL cells isolated from two healthy donors and two CLL patients, respectively, were exposed to the indicated concentrations of PEITC for 72 h and cell viability was monitored by MTT assay. (A) Cell viability curves of normal lymphocytes (blue symbols) and CLL cells (red symbols). (B) The concentration required to cause a loss in cell viability by 50% (IC50) for each sample was calculated from the respective viability curve.





  • Figure S2: Cytotoxic effect of PEITC, F-ara-A, and their combination in CLL cells (JPG, 83.1 KB) -
    (A) Assay of cell death induced by F-ara-A (20 µM, 48 h), PEITC (5 µM, 4h), or the combination of the two compounds (F-ara-A 44 h followed by addition of PEITC for another 4 h) in primary CLL cells. Cell death was analyzed using annexin-PI double staining followed by flow cytometry analysis. Representative dot plots of a patient sample are shown. (B) Quantitative analysis of the combined cytotoxic effect of F-ara-A and PEITC in CLL cells isolated from 4 blood samples, using the experimental conditions described in (A). The expected viable cell fraction (%) was calculated by multiplying % viable cells in the F-ara-A-treated sample with the % viable cells in the PEITC-treated sample. The combined drug effect is considered additive if the observed viable cell fraction from the experiments is equal to the expected value. If the observed value were less than the expected value, the combined drug effect is considered as more than additive.
