(A) Xanthohumol does not inhibit TNF-induced AP-1 activation in vivo. KBM-5 (2 × 106 cells/well) were treated with different concentrations of xanthohumol for 4 h and then treated with 0.1nM TNF for 30 min, prepared NE and the activation of AP-1 was analyzed by EMSA (15 µg/well for EMSA). (B) Xanthohumol does not inhibit TNF-induced SP-1 activation in vivo. KBM-5 (2 × 106 cells/well) were treated with different concentrations of xanthohumol for 4h and then treated with 0.1nM TNF for 60 min, prepared NE and the activation of SP-1 was analyzed by EMSA. (C) Xanthohumol does not inhibit TNF-induced AP-1 activation in vitro. KBM-5 (2 × 106 cells/well) were treated with 0.1nM TNF for 30 min, prepared NE and incubated with different concentrations of xanthohumol for 30 min at 37°C, and the activation of AP-1 was analyzed by EMSA. (D) Xanthohumol does not inhibit TNF-induced SP-1 activation in vitro. KBM-5 (2 × 106 cells/well) were treated with 0.1nM TNF for 60 min, prepared NE and incubated with different concentrations of xanthohumol for 30 min 37°C, and the activation of SP-1 was analyzed by EMSA.
