A culture of CTL RP1 with >95% LRH-tetramer+ cells was compared to a culture of CTL TB1 with ~33% LRH-1–tetramer+ cells at an E:T ratio of 1:3 corrected for the tetramer+ T cell content. Depicted are duplicate ELISA measurements of IFN-γ production by each CTL after overnight stimulation. We observed that IFN-γ production by CTL TB1 was slightly higher than by CTL RP1, suggesting a somewhat higher avidity of CTL TB1 for LRH-1+ target cells. (B) Overnight stimulation of CTL with LRH-1+ EBV-LCL showed high levels of IFN-γ production, which was similar for CTL RP1 and TB1 compared with HLA-B7–specific CTL KOR18.
