(A) The flow cytometric gating strategy employed in order to positively identify CLL cells for Mcl-1 quantification. Both R1 and R2 were forward gated onto the histograms for Mcl-1 expression. (B) Representative histograms from eight patient samples (#15, 26, 38, 73, 84, 93, 107 and 136) in which the quantitative fluorescent values for Mcl-1 expression are given as molecules of equivalent soluble fluorochrome (MESF) as derived from the geometric mean of the histograms. (C) Western blot analyses from the same eight patients. Bands were quantified by scanning densitometry and comparison of the MESF values (B) and the Mcl-1:Actin ratios (C). Concordance between the two methodologies is shown (D).

Serial samples from eleven patients were collected over a period of up to 48 months in order to determine whether the expression of Bcl-2, Bax and Mcl-1 were modified by time, clinical progression or treatment. (A) There was a trend towards increased Bcl-2 expression in the same patient over time and this was most marked in patients who had received chemotherapy. (B) Bax expression was often reduced and again this was most pronounced in patients who had received chemotherapy. (C) In contrast, Mcl-1 expression remained largely unchanged over time in all eleven patients.
