Hepatitis C virus association with peripheral blood B lymphocytes potentiates viral infection of liver-derived hepatoma cells
Blood Stamataki et al. 113: 585

Supplemental materials for: Stamataki et al

Files in this Data Supplement:

  • Figure S1. Effect(s) of stimulation on B-cell association with JFH-1 (JPG, 55.6 KB) -
Untreated and CD40L/IL-4 stimulated L3055-Bcl-2 and KEM cells were incubated with JFH-1 for 2h at 37°C, unbound virus removed by washing and cells analyzed for HCV RNA by RT-PCR (A) or trans-infection of Huh-7.5 (B). Error bars represent the SD from 3 or 4 replicate infections.

  • Figure S2. Comparative infectivity of cell-free and B-cell–associated JFH-1 (JPG, 26.8 KB) -
Infectivity of cell-free (open circle and dotted line) and CD40L/IL-4–stimulated L3055-Bcl-2 (filled circle) and PBMC derived B-cell–associated JFH-1 (BD1-open, BD2-filled squares) at 37°C for different time periods. Infectivity is expressed relative to cells co-cultured with Huh-7.5 immediately following virus loading. The relative infectivity retained by primary B cells following incubation at 37°C for 2.5 and 4.5 hours is significantly higher than that retained either by cell-free virus or L3055-Bcl-2 cells (Dunn’s test, p=<0.05).
