Mononuclear cells were isolated from peripheral blood and cultivated in EGM-2MV BulletKit medium for 4 days. (A) Human EPCs stained with Dil-acLDL (red); (B) Human EPCs stained with SR-BI antibody (green); (C) Merged view of Dil-acLDL and SR-BI double positive cells. Scale bar represents 100 µm.

Cholesterol lipoprotein profiles obtained by fast performance liquid chromatography gel filtration of 100 µl plasma obtained at day 0 (F), day 14 (E), and day 35 (H) after gene transfer with AdA-I or Adnull in C57BL/6 mice 4 weeks after transplantation with SR-BI+∕+ or SR-BI−∕− bone marrow. Fractions 12–32 correspond to non-HDL sized particles whereas fractions 33–45 correspond to HDL sized particles.

Bar graph showing the number of migrated EPCs in modified Boyden chambers. After 7 days of culture, Sca-1 positive bone marrow EPCs isolated from wild-type or SR-BI−∕− mice were seeded in the upper chamber. The lower chamber was supplemented with either HDL (100 µg/ml) or an equivalent amount of bovine serum albumin and the number of migrated cells per microscopy field was quantified after 5 hours (n=4 for each group).

-
Representative sections of Mac-3 positive macrophages at day 21 after transplantation in control (A, C) and AdA-I (B, D) treated C57BL/6 mice with SR-BI+∕+ (A–B) or SR-BI−∕−(C–D) bone marrow. Scalebar represents 100 µm.
