Resting platelets do not generate microparticles (JPG, 46.4 KB) - Platelet-rich plasma (PRP) was kept resting by incubation with lepirudin as described in the Materials and Methods. (A) At the indicated times, samples of PRP were analyzed for P-selectin surface expression by flow cytometry. P-selectin surface expression in a sample exposed to 1 U/ml thrombin plus 50 µg/ml collagen (T+C) was also measured for comparison. Error bars represent the S.D. of 4 independent experiments. (B) Cleavage of filamin A in samples of resting PRP incubated for the indicated times was assessed by immunoblot analysis. (C) PRP incubated for the indicated time points was analyzed for microparticle production by flow cytometry. Microparticle formation from a sample exposed to 1 U/ml thrombin plus 50 µg/ml collagen (T+C) was measured for comparison. Data represent microparticles as a percent of total events (microparticles and platelets). Error bars represent the S.D. of 4–6 independent experiments. Video 1. Richardson Technology Microscopy of microparticle formation by cultured mouse megakaryocytes (AVI, 1.87 MB) - Evaluation of day 3 megakaryocytes using Richardson Technology Microscopy demonstrates microparticle formation along the surface of the plasma membrane.
