Naïve CD4+ T cells were purified from the peripheral blood by negative selection using the EasySep Human Naïve CD4+ T-Cell Enrichment Kit (Stem Cell Technologies) and stained with CFSE. Cells were cultured for 7 days in the presence or absence of IL-7. At Day 1 and at Day 7 cells were harvested and stained using monoclonal antibodies against CD4, CD45RA, and CD31. The percentage of CFSE negative was quantified following gating on CD4+CD45RA+ population and subsequent gating on CD31+ or CD31− subsets. Unstimulated cells were used as an additional control, and results obtained at Day 7 were similar to the ones at Day1 (data not shown).
