This heatmap depicts relative protein values obtained from antibody microarrays after removal of batch effects due to three separate analyses. Samples from 21 GVHD− patients (left panel) and 21 GVHD+ patients (right panel) are represented. Only the antibodies giving the 35 smallest p-values for differences between GVHD+ and GVHD− patient plasma are shown. P-values compare GVHD+ and GVHD− samples.

-
Scores were derived from diagonal linear discriminant analysis (DLDA) of levels measured by ELISA in the 42 samples of the discovery set. We used the N assays with smallest p-values for comparing GVHD+ versus GVHD− samples (as determined by the remaining samples), as N varies between 1 and 15 assays. Area under the Receiver Operating Characteristic (ROC) curve (black squares) and p-value from one-sided Rank-Sum Test (open diamonds) as determined by LOOCV are displayed as a function of the number of assays used.

- Figure S3. Optimal ROC curve from LOOCV in the discovery set (JPG, 36.9 KB)
-
ROC curve obtained by LOOCV of diagonal linear discriminant scores for the 42 discovery samples using the eight biomarkers with the smallest p-values as measured by ELISA. The AUC was 0.94.

- Figure S4. HGF and IL-2Rα levels and target organ at onset (JPG, 42.2 KB)
-
Mean levels of HGF and IL-2Rα are represented on the y axis as a function of target organ at onset. (A) Mean HGF level is significantly elevated in visceral only and skin/visceral GVHD as compared to skin only GVHD * (ANOVA adjusting for the other panel biomarker). (B) Mean IL-2Rα level is significantly higher in skin and skin/visceral GVHD than in visceral only GVHD * (ANOVA adjusting for the other panel biomarker).
