(A) Alignment of A20, c2B8, and hA20 VH chains. Dots indicate homology to A20; green text, framework residues; boxed region with red text, CDR. (B) Alignment of A20, c2B8, and hA20 Vk chains. Dots indicate homology to A20; green text, framework residues; boxed region with red text, CDR.

- Figure S2. Binding characteristics of veltuzumab and rituximab were determined by binding the 125I-labeled mAbs to Raji cells (JPG, 69.4 KB)
-
Direct cell surface saturation binding and Scatchard plot analysis (insert). Closed triangle: veltuzumab; open circle: rituximab. Bmax and Kd were determined by non-linear regression analysis using a one-site binding model with Prism software. These results are representative of one of three repeated experiments.

- Figure S3. Effects of veltuzumab and rituximab on proliferation of non-Hodgkin’s lymphoma cell lines (JPG, 47.8 KB)
-
Anti-proliferative effects were assessed by MTT cytotoxicity assays. Cells were cultured with the mAbs with or without a second antibody for crosslinking. White bars, no second antibody; gray bars with GAH second antibody; error bars, SD.

- Figure S4. PK of veltuzumab in normal mice (JPG, 26.8 KB)

Veltuzumab (150 µg) was administered to naïve Swiss-Webster mice either via the i.p. or s.c. route. Animals were bled over a 14-day period and serum was assayed for veltuzumab concentrations as described in the Methods section. (N=6).
