Allogeneic T lymphocytes do not survive in unmanipulated or minimally lymphodepleted hosts (JPG, 39.8 KB) - Pmel-1b/d cells were generated from pmel-1-BALB/c-B6 F1 mice and transferred into non irradiated C57BL/6 (B6 NO RAD) or into C57BL/6 that received a dose of 5 Gy total body irradiation the day before (B6 5 Gy) or into Rag1−/− mice (Rag1−/−). After 5 or 10 days, mice were sacrificed and the spleens were analyzed for the presence of allogeneic cells expressing the H-2 Dd molecule. Results are representative of 2 independent experiments. C57BL/6 mice bearing 10 day old B16 tumors were irradiated with 9 Gy TBI and injected with 106 allogeneic pmel-1b/d cells, vaccinia virus encoding hgp100, and exogenous rhIL-2 (PVI) (JPG, 51.9 KB) - All groups received syngeneic bone marrow transplant with 106 C57BL/6 (H-2b/b) the day after the transfer of the effector cells. After 21 days mice were sacrificed and the spleens, draining lymph nodes (DLN) or tumor and were analyzed by flow cytometry for the presence of the transferred cells. The dot-plots show the percentage of CD8+ H-2d+ cells. Results are representative of two independent experiments. Figure S3.
