  • Figure S1. BCR-ABL activates high fidelity HDR repair in DRGFP reporter containing BaF3 cells (JPG, 282 KB) -
    (A) The DRGFP reporter for measuring HDR contains a full-length non-functional GFP gene (SceGFP) interrupted by an I-SceI site. Repair by HDR of the DSB generated by I-SceI using the downstream GFP sequence (iGFP) results in a functional GFP gene. (B) BaF3.BCR-ABL cells stably expressing the HDR reporter were used to measure repair fidelity in response to I-SceI induced DSBs. Genomic DNA from clonal populations of GFP-positive BaF3.BCR-ABL cells containing the reporter was isolated to assess the repaired GFP region by PCR (flanking primers indicated by arrows) and sequencing (partial DNA sequence shown).
