T-cell and B-cell immune function assessment The total peripheral blood lymphocyte count and various lymphocyte subsets were evaluated using flow cytometry (CD3+, CD3+CD4+, CD3+CD8+, as well as CD19/CD20+ B cells, CD3−CD16+ NK cells). Since 2000, we have also analyzed naive CD31+CD45RA+ T-cell counts within the CD3+CD4+ subset as an indicator of the naive T-cell population; this parameter was found to correlate strongly with the determination of T-cell receptor excision circles (+).28 In vitro lymphocyte proliferation induced by mitogens and antigens was evaluated by 3Hthymidine uptake. The serum levels of IgG, IgA and IgM were measured by nephelometry. Serum antibodies to polioviruses and tetanus and diphtheria toxoids were measured using enzyme-linked immunosorbent assays after booster immunization in some patients.
