5 × 104 or 5 × 106 LN CD4+ T cells from CD45.1 C57BL/6 mice were purified, labeled with CFSE and transferred into CD45.2 C57BL/6 CD3ε−∕− mice. Seven, 14 and 28 days after transfer, pooled LNs and spleen were analyzed for Foxp3 expression. (A) Fluorescence Foxp3/FSC dot plots are shown for CFSE− CD45.1+ CD4+ CD8− CD3+ recovered T cells. (A) LN CD4+ T cells from CD45.1 C57BL/6 mice were purified and naive CD4+ T cells were electronically sorted on the basis of their nonexpression of CD25 and their low or absent expression of CD44. Fluorescence dot plots show CD45RB and CD69 expression on sorted CD25− CD44−∕low CD4+ (top) or CD25−∕low CD44high CD4+ (bottom) T cells. (B) 5 × 103 purified CD25− CD44−∕low CD4+ or CD25−∕low CD44high CD4+ T cells from CD45.1 C57BL/6 mice were injected into CD45.2 C57BL/6 CD3ε−∕− mice. Absolute numbers of recovered CD45.1+ CD4+ CD8− TCR+ cells are shown one month after transfer. ns, not significant.
