Lymphopenia-induced spontaneous T-cell proliferation as a cofactor for autoimmune disease development
Blood Le Campion et al. 114: 1784

Supplemental materials for: Le Campion et al

Files in this Data Supplement:

  • Figure S1. The frequency of regulatory T cells among CD4+ T cells undergoing spontaneous proliferation increases with time (JPG, 268 KB) -
5 × 104 or 5 × 106 LN CD4+ T cells from CD45.1 C57BL/6 mice were purified, labeled with CFSE and transferred into CD45.2 C57BL/6 CD3ε−∕− mice. Seven, 14 and 28 days after transfer, pooled LNs and spleen were analyzed for Foxp3 expression. (A) Fluorescence Foxp3/FSC dot plots are shown for CFSE CD45.1+ CD4+ CD8 CD3+ recovered T cells. (B) Proportion of CFSE CD45.1+ CD4+ CD8 CD3+ T cells expressing Foxp3. (C) Absolute numbers of recovered CFSE CD45.1+ CD4+ CD8 CD3+ T cells expressing (filled) or not (opened) Foxp3. Results are from two independent experiments.

  • Figure S2. Spontaneous proliferation of naive or activated/memory CD4+ T cells in C57BL/6 mice (JPG, 162 KB) -
    (A) LN CD4+ T cells from CD45.1 C57BL/6 mice were purified and naive CD4+ T cells were electronically sorted on the basis of their nonexpression of CD25 and their low or absent expression of CD44. Fluorescence dot plots show CD45RB and CD69 expression on sorted CD25 CD44−∕low CD4+ (top) or CD25−∕low CD44high CD4+ (bottom) T cells. (B) 5 × 103 purified CD25 CD44−∕low CD4+ or CD25−∕low CD44high CD4+ T cells from CD45.1 C57BL/6 mice were injected into CD45.2 C57BL/6 CD3ε−∕− mice. Absolute numbers of recovered CD45.1+ CD4+ CD8 TCR+ cells are shown one month after transfer. ns, not significant. Each point represents an individual mouse and horizontal lines represent the mean number of CD4+ T cells recovered from positive mice.
