- Figure S1. Post sorting purity of CD44low naïve T cells (JPG, 8.61 KB)
-
Purified lymph node T cells were stained with anti-CD44 and CD44low naïve T cells were subsequently sorted using a FACSAria sorter. A typical post sorting profile of CD4 and CD8 T cells is shown.

- Figure S2. Total cell recovery after T-cell transfer (JPG, 38.1 KB)
-
Groups of Rag1−∕− and TCRβ−∕− mice were transferred with 1 × 106 naïve T cells. Mice were sacrificed every week after the transfer, and total donor cell recovery was determined by FACS analysis. Shown are the mean ± SD (n=3). *, p<0.05; **, P<0.01; ***, P<0.001.

- Figure S3. γδ T-cell depletion by anti-Thy1.2 Ab in TCRβ−∕− mice (JPG, 13.7 KB)
-
TCRβ−∕− mice were injected i.p. with anti-Thy1.2 or control rat IgG Ab (500 µg/mouse on days −2, 1 and 3). γδ T-cell depletion was examined at 7 days post injection.

- Figure S4. Donor CD4 and CD8 T-cell recovery in γδ T cells depleted TCRβ−∕− mice at 4 weeks post transfer (JPG, 11.7 KB)
-
Mice were treated with 500 µg control Ig (●) or anti-Thy1.2 mAbs (○) on days −2, 1 and 3. At day 0, 1 × 106 Thy1.1 naïve T cells were transferred into these TCRβ−∕− mice. The recipient mice were sacrificed 4 weeks after T-cell transfer and the recovery of Thy1.1+ cells in the spleen, the peripheral LN, and the mesenteric LN were calculated by FACS analysis. The results are representative of 5 individual mice per group from two separate experiments. Shown are the mean ± SD. **, P < 0.01.

- Figure S5. IL-15Rα expression on CD11c+ DCs (JPG, 23 KB)
-
Groups of Rag1−∕−, TCRβ−∕−, and anti-Thy1.2 Ab treated TCRβ−∕− mice were sacrificed and LN, mLN, and spleen cells were harvested. The cells were immediately stained for CD11c and IL-15Rα. The mean fluorescence intensity is shown.

- Figure S6. Impaired CD8 T-cell expansion after IL-15−∕− BMDC transfer (JPG, 40.1 KB)
-
TCRβ−∕− mice were transferred with 1 × 106 naïve Thy1.1 T cells together with BMDCs derived from either wild type or IL-15−∕− mice. Mice were sacrificed 14 days after transfer. FACS dot plots of the indicated lymphoid tissues are shown (representative of 3~4 individually tested mice).
