Reporter expression experiments To understand the pathways involved in Gaucher disease pathophysiology, we analyzed the regulatory elements of co-regulated genes using a bioinformatic tool (DiRE). Noteworthy among the most frequent supposed regulatory elements are those recognized by STAT1 and SRF. To validate this insight, we studied reporter gene expression in CBE treated human 293T cells. Cignal Finder Immune Response Reporter Array plates from SABioscience were used to reverse transfect 293T cells using Polyfect (Qiagen) according to the manufacturer’s instructions. Twelve hours after the transfection, CBE (100 µM) was applied to the cells for 48 hours before firefly luciferase (reporter gene) and renilla luciferase (internal control) activities were assessed using the Dual Luciferase assay (Promega, Madison, WI, USA) and a Fluorostar Optima plate reader (BMG Labtech, Chicago, IL).
