Shown at 2 months and 1 year post-transplant are percent Vβ expression on splenic T cells from AKR/J → BALB.K (H2k) mixed chimeras for (A) AKR/J donor-derived Thy1.1-positive T cells and (B) BALB.K host-derived Thy1.2-positive T cells. Control AKR/J mice delete Vβ5 and Vβ6, whereas control BALB.K mice delete Vβ4 and Vβ5. Values are median and error bars indicate standard deviation.

(A) Splenocytes were harvested from mice immediately after sublethal TBI, cultured in cIMDM supplemented with 20% FBS, fixed with ethanol at indicated time points, and stained with propidium iodide (PI) for FACS analysis. Shown are histograms of PI fluorescence with gate over subdiploid G0∕1 peak. (B) Mice (n=3 per strain) were given sublethal TBI and sacrificed at Day+3. Shown are median percent viable cells in spleen, cervical lymph node, thymus and bone marrow, and (C) representative FACS analysis of thymocytes for CD4-PE and CD8-FITC.

Interval mapping was performed in R/qtl using the method of Broman36 analyzing separately for the log-transformed T-cell chimerism trait value, conditional on attainment of engraftment, results of which are shown as LOD (µ) (blue line); for the binary trait value, shown as LOD (p) (red line); and a combined analysis for both parameters, shown as LOD (p,µ) (black line). Note that LOD (p) is identical to the LOD plot, shown in Fig. 6C, for interval mapping of the binary engraftment trait. Horizontal dotted lines indicate significant (p < .05) LOD threshold levels for LOD (p,µ) (black dotted line), LOD (p) (red dotted line), and LOD (µ) (blue dotted line), determined by 10,000 permutation tests.
