CD8{alpha}+ mouse spleen dendritic cells do not originate from the CD8- dendritic cell subset

Although previous studies had indicated that the CD8{alpha}- and CD8{alpha}+ subtypes of murine dendritic cells (DC) differ in immediate origin, a recent study found that intravenous transfer of CD8{alpha}- DC led to CD8{alpha}+ DC in the spleen several days later, suggesting to a direct precursor-product relationship. We have repeated these experiments with a balance-sheet approach. We find that although a few CD8{alpha}+ DC can be generated in such experiments, this is a rare event and could be due to a contaminant precursor. The vast majority of the immediate precursors of CD8{alpha}+ DC are cells that lack the phenotype of a recognisable DC. CD8{alpha}- DC and CD8{alpha}+ DC are not precursor-product related, although these sublineages may be connected further upstream.
